چکیده
Abstract Background Patients suffering from IBD frequently suffer chronic visceral pain, severely affecting quality of life. To better understand factors impacting pain signaling by enteric nervous system (ENS) neuronal cells, we analyzed the infiltration myenteric plexus (MP) ENS inflammatory cell populations. In detail, characterized (i) immune populations infiltrating MP, (ii) neurotransmitter expression MP cells and (iii) survival in Crohn’s disease (CD) ulcerative colitis (UC). Methods We included 12 UC nine CD patients that received colectomies or ileocecal resections, respectively, 10 control with surgeries for non-inflammatory reasons. FFPE material was used immunohistochemistry analysis MP. Immunostaining performed detection neurons (beta-III tubulin, PGP9.5), T-cells (CD3, CD4, CD8 FOXP3), B-cells (CD20), monocytes (CD68, CD163) apoptosis (Annexin V) neurotransmitters CGRP substance P. Intraganglionic periganglionic were determined conventional LSM confocal microscopy. Results CD3+ CD8+ intraganglionic (CD3+: 1.7 ± 0.6 cells/10.000 µm²; CD8+: 4.8 1.1 µm²) found to be increased compared 0.2 0.1 µm², p=0.04; 0.4 p=0.006), Figure 1. CD20+ CD68+ significantly as well (CD20+ CD: 0.8 controls: 0 µm, p=0.02), 12.3 3.5 p=0.001). cytotoxic area UC. 1.6-fold detected p=0.01. Substance P 0.7-fold reduced MP-cell p=0.048. 2 shows representative immunostainings two per condition. Conclusion conclusion, infiltrations are comprised CD3+, B-cells. Specifically periganglionically, The is CD-MPs whereas levels UC-affected MPs revealed higher apoptosis.

In Silico Prediction of B-Cell and T-Cell Epitopes of Protective Antigen of Bacillus anthracis in Development of Vaccines Against Anthrax
Protective antigen (PA), a subunit of anthrax toxin from Bacillus anthracis, is known as a dominant component in subunit vaccines in protection against anthrax. In order to avoid the side effects of live attenuated and killed organisms, the use of linear neutralizing epitopes of PA is recommended in order to design recombinant vaccines. The present study is aimed at determining the dominant epi...
متن کاملMyenteric plexus in Hirschsprung's disease.
The absence of ganglion cells and the presence of abnormal bundles of unmyelinated fibres in the affected segment in Hirschsprung's disease was first well described by Whitehouse and Kernohan (f948). However, previous authors who have studied this problem have used standard transverse sections of the gut to demonstrate the abnormalities but these give a very inadequate view of the plexus which ...
متن کاملEffects of berberine on the secretion of cytokines and expression of genes involved in cell cycle regulation in THP-1 monocytic cell line
Objective(s): Current acute myeloid leukemia (AML) therapeutic strategies have irreversible side-effects. Berberine (BBR) is an isoquinoline alkaloid, which has been known as an aryl hydrocarbon receptor (AhR) ligand. AhR is a cytoplasmic receptor, which is involved in the regulation of cellular and immune responses. Here, we investigated the expression profile of genes involved in the cell cyc...
متن کاملB and T Lymphocyte Attenuator is a Target of miR-155 during Naive CD4+ T Cell Activation
Background: MicroRNA-155 (miR-155) is upregulated during T cell activation, but the exact mechanisms by which it influences CD4+ T cell activation remain unclear. Objective: To examine whether the B and T lymphocyte attenuator (BTLA) is a target of miR-155 during naïve CD4+ T cell activation. Methods: Firefly luciferase reporter plasmids pEZX-MT01-wild-type-BTLA and pEZX-MT01-mutant-BTLA were ...
متن کاملMir-55 inhibition can reduce cell proliferation and induce apoptosis in Jurkat (Acute T cell Leukemia) cell line
Background MicroRNAs are small and non-coding RNA molecules with approximately 22 nt in length that cause inhibition of translation or degradation of mRNA. MiR-155 is a kind of molecule with different functions, such as its role in proliferation, apoptosis, inflammation, differentiation, and immunity. One of its best known functions is apoptosis that affects on caspase-3 activity. The main aim...
